ABSTRACT

Background: Elderly individuals diagnosed with high-grade gliomas frequently experience unfavorable outcomes. We aimed to design two web-based instruments for prognosis to predict overall survival (OS) and cancer-specific survival (CSS), assisting clinical decision-making. Methods: We scrutinized data from the SEER database on 5,245 elderly patients diagnosed with high-grade glioma between 2000-2020, segmenting them into training (3,672) and validation (1,573) subsets. An additional external validation cohort was obtained from our institution. Prognostic determinants were pinpointed using Cox regression analyses, which facilitated the construction of the nomogram. The nomogram's predictive precision for OS and CSS was gauged using calibration and ROC curves, the C-index, and decision curve analysis (DCA). Based on risk scores, patients were stratified into high or low-risk categories, and survival disparities were explored. Results: Using multivariate Cox regression, we identified several prognostic factors for overall survival (OS) and cancer-specific survival (CSS) in elderly patients with high-grade gliomas, including age, tumor location, size, surgical technique, and therapies. Two digital nomograms were formulated anchored on these determinants. For OS, the C-index values in the training, internal, and external validation cohorts were 0.734, 0.729, and 0.701, respectively. We also derived AUC values for 3-, 6-, and 12-month periods. For CSS, the C-index values for the training and validation groups were 0.733 and 0.727, with analogous AUC metrics. The efficacy and clinical relevance of the nomograms were corroborated via ROC curves, calibration plots, and DCA for both cohorts. Conclusion: Our investigation pinpointed pivotal risk factors in elderly glioma patients, leading to the development of an instrumental prognostic nomogram for OS and CSS. This instrument offers invaluable insights to optimize treatment strategies.

ABSTRACT

Winter flooding of harvested rice fields is a typical cropping system in mountainous areas, which emits considerable amounts of CH4. Plastic film mulching cultivation is recognized as an important rice cultivation practice in paddy field for water-saving irrigation. However, the effects of these managements on CH4 emissions in paddy soil and the underlying microbial mechanism are unclear. A field experiment was carried out with the application of winter drainage followed by traditional rice cultivation (WD), winter drainage followed by plastic film mulching cultivation (MC), as well as winter flooding followed by traditional rice cultivation (WF) as control in hilly paddy fields. We investigated the CH4 emissions, functional (CH4 production rate, 13C isotope) and structural (abundance, structure) responses of soil methanogenic archaeal and fermenting bacterial communities during rice season. Shifting the fields from WF into WD and MC substantially mitigated CH4 emissions by 62.3% and 59.2%, respectively, paralleled with the enhancement of soil Eh and the reductions of soil DOC content. Compared with WF, WD and MC both significantly decreased CH4 production rates and the copy numbers of mcrA gene. Moreover, an increasing contribution of hydrogenotrophic methanogenesis (from 30.7% to 50.0%) to total CH4 production was observed during the conversion from WF to MC under an anaerobic incubation, paralleled with the decreased acetate content and increased δ13C values of acetate-methyl and total acetate. The communities of methanogenic archaea and fermenting bacteria strongly responded to the shift from WF to WD, while MC only showed significant effects on the methanogenic archaeal communities. Compared with WF, WD and MC significantly increased the relative abundance of Methanothrix, Methanosarcina and Methanocella, while those of Methanoregula, Massilia and Geobacter were decreased. The co-occurrence networks showed that WD and MC induced the loss of mixed methanogenic fermentation modules, indicating the decrease in functional biodiversity and redundancy of fermenting bacterial and methanogenic archaeal communities.The findings suggest that WD and MC approach mitigate CH4 emission by regulating the function and structure of methanogenic archaeal and fermenting bacterial communities in paddy soil, which represent the effective management strategies considering the water availability and CH4 mitigation in paddy-field agriculture.

ABSTRACT

Hydrothermal carbonization (HTC) technology can potentially be used to safely and sustainably utilize kitchen waste (KW). However, the characteristics of HTC solid products (hydrochar) and aqueous products (HAP) based on different types of KW have not yet been clarified. Here, four types of KW, cellulose-based (CL), skeleton-based (SK), protein-based (PT), and starch-based (ST) KW, were used for HTC at 180 °C, 220 °C, and 260 °C. The basic physicochemical properties and structures of hydrochars and HAP were analyzed, and the effects of different hydrochars on rice growth were characterized. HTC decreased the H/C and O/C of KW. All hydrochars were acidic (3.12 to 6.78) and the pH values increased with the HTC temperature, while high HTC temperature reduced the porosity of hydrochars. HTC promoted the enrichment of total carbon (up to 78.1 %), total nitrogen (up to 62.6 %), and total phosphorus (up to 171.6 %) in KW. More carbon (60.7-88.0 %) and nitrogen (up to 87.4 %) were present in the hydrochars than in the HAP. The relative content of C1s increased and O1s decreased in CL and ST hydrochars as the HTC temperature increased, while the opposite pattern was observed for SK and PT hydrochars. The dissolved organic matter (DOM) of different hydrochars and HAP were mainly humus-like substances. The biodegradability of the DOM in HAP was often higher than the corresponding hydrochar, and their DOM biodegradability increased with the HTC temperature. The content of heavy metals from different hydrochars did not exceed the relevant thresholds of fertilizer standards. Rice grain yield increased by 3.7-11.1 % in the hydrochar treatments without phosphate fertilizer addition compared with the control treatment. The results of this study provide new theoretical and empirical insights into the potential for HTC technology to be used for the recycling of KW and its products in the agricultural environment.

ABSTRACT

Al-doped spinel LiNi0.5Mn1.5O4 materials with different sites and contents were synthesized by rapid precipitation combined with hydrothermal treatment and calcination. The roles of Al on structural stability and electrochemical performance were studied by utilizing a series of techniques. XRD patterns indicated lower ion diffusion and no impure phased in doped samples. FT-IR and CV results reveal that Al-doped materials possess a Fd3̄m space group with increased disorder and increasing amounts of Mn3+. SEM and TEM equipped with EDS were used to characterize the regular morphology accompanied by a complete crystal structure and homogeneous distribution of elements. The Al content at the Ni, Mn, and Ni/Mn sites was optimized to be 5%, 3% and 5% (in total), respectively. The cycling stability was considerably enhanced at an ambient temperature (25 °C) and high temperature (55 °C). A typical Al dual-doped sample at Ni/Mn sites with 5% content delivered a reversible capacity of 113.5 mA h g-1 after 200 cycles at 0.5C. The discharge capacity at 5, 10 and 20C was 127.3, 125.5 and 123.1 mA h g-1, respectively. The discharge capacity remained at 126 mA h g-1 after 50 cycles (55 °C, 0.5C). Subsequent EIS and analytical results of the cycled electrode showed improved structural stability with a lower resistance, stable cathode/electrolyte interface, and reduced dissolution of Mn. These data further demonstrated the feasibility and reliability of preparing high-performance spinel LiNi0.5Mn1.5O4 cathode materials by doping with a suitable amount of Al.

Kidney transporters are involved in the secretion and reabsorption of endogenous and exogenous molecules. Numerous factors may influence their expression and affect drug disposition, efficacy and toxicity. The present study aimed to examine the development­ and age­associated variations in primary renal transporters in rats, including 6 uptake transporters: Organic anion transporter (OAT) 1 and 3, organic cation transporter (OCT) 1, 2 and 3 and organic anion­transporting polypeptide (OATP) 4C1, and 6 efflux transporters: Multidrug resistance protein 1 (MDR1), breast cancer resistance protein (BCRP), multidrug resistance­associated protein (MRP) 2 and 4, and multidrug and toxin extrusion protein (MATE) 1 and 2­K. Kidneys from male Sprague Dawley rats during development (­2, 1, 7, 14 and 21 days), maturation (28, 35 and 60 days) and aging (180, 540 and 850 days) were collected and total RNA was extracted, purified and subjected to reverse transcription­quantitative polymerase chain reaction analysis. Total proteins were extracted for western blot analysis. OAT1 and 3, OCT1, BCRP, MRP2 and 4 and MATE2­K expression levels were low in fetal kidneys, increased gradually following birth and markedly increased on maturation and adulthood. High levels were maintained until 850 days. OCT2, OATP4C1, Mdr1b and MATE1 expression levels were low in fetal kidneys, increased gradually following birth, and increased markedly on weaning, maturation and adulthood; however, levels were decreased on aging. OCT3 mRNA expression levels were low in fetal and newborn kidneys, and had two peaks at 35 and 850 days. The selected OAT1 and 3 and MDR1 protein expression levels revealed a similar expression pattern. Thus, kidney transporter expression is affected by ontogeny and aging, which may impact drug and toxicant disposition in children and the elderly.
